Siglec-3 Background

Myeloid cell surface antigen CD33 is also known as SIGLEC3, Siglecs (sialic acid binding Ig­like lectins) and GP67, is a single-pass type I membrane protein which belongs to the immunoglobulin superfamily and SIGLEC (sialic acid binding Ig-like lectin) family. Human CD33 / Siglec-3 cDNA encodes a 364 amino acid (aa) polypeptide with a hydrophobic signal peptide, an N-­terminal Ig-­like V­-type domain, one Ig­-like C2-­type domains, a transmembrane region and a cytoplasmic tail. CD33 / Siglec-3 usually considered myeloid-specific, but it can also be found on some lymphoid cells. In the immune response, CD33 / Siglec-3 may act as an inhibitory receptor upon ligand induced tyrosine phosphorylation by recruiting cytoplasmic phosphatase(s) via their SH2 domain(s) that block signal transduction through dephosphorylation of signaling molecules. CD33 / Siglec-3 induces apoptosis in acute myeloid leukemia.

    While transducing primary cells, I was running into a staining issue with biotinylated-rCD33. A colleague of mine had used rCD33 labelled with PE before and had success with a similar experiment. I decided to order from Acrobio Systems due to the recommendations and high praise from several colleagues. Upon staining, I saw great fluorescence, increased events caught, and higher transduction efficiencies. I will definitely be using this product again!

    We purchased this reagent for use in detecting CD33 CAR expression and it worked as advertised. The PE signal gave great separation by flow cytometry and allowed for the clear detection of CAR expression on the surface of our cells._x000D_ _x000D_ The product page states that the CD33 PE reagent is incompatible with streptavidin-biotin detection systems. When we tried to co-stain with another biotinylated reagent, subsequent application of streptavidin-APC resulted in high non-specific APC staining. This could be avoided by staining with CD33 PE after all other staining steps have been completed.
